Can it strip multicore cable in one pass? Or do I need to break the larger cable down into their individual cores for a second pass? I mostly need to strip 5 core 4mm heavy duty flex and orange Circular power cable.

No, it cannot strip multi-core cables in one go, you need to strip it a second time.
